CULTIVATION WITH CHLORELLA VULGARIS UTEX 1813 USING AGROINDUSTRIAL SUBSTRATES.

SUMMARY:

The research proposal is aimed at the co-cultivation of the filamentous fungus Mucor Circinelloides and the microalgae Chlorella vulgaris for the formation of rich biomass and the production of active biomolecules, from secondary metabolism, using renewable substrates (crude glycerol as a source of organic carbon and as a source of corn-cine nitrogen), through the circular bioeconomy. In this first stage, the filamentous fungus Mucor circinelloides UCP 0018 was used to investigate the influence of carbon and nitrogen sources on the biotechnological production of biosurfactant and lipid biomass. The agro-industrial residues were used: crude glycerol and corn steep liquor for the formulation of economic and sustainable culture media, supplemented KH2PO4, MgSO4, FeSO4.7H2O, MnSO4 e ZnSO4. A full factorial design 22 was use to investigate the influence of these variables through a submerged fermentation at 150 rpm, at 28°C, pH 5.5 for 96h. To investigate the biosurfactant produced, tests were carried out to determine the surface tension, emulsification index, dispersion and critical micellar concentration and, finally, the characterization of the biosurfactant through infrared and determination of the ionic charge, were also determined the protein content , lipids and carbohydrates. For the investigation of biomass, its yield was analyzed and lipids were extracted.The analyzes showed that the production of fungal biosurfactant and lipid-rich biomass was positively influenced by the carbon and nitrogen sources tested. of 29.7 mN/m, it reached an emulsification index of more than 98% and a dispersion in motor oil of 28.26 cm². As for surface tension, the biosurfactant produced showed thermal stability (20°C to 80°C), at different pH (2 to 8) and NaCl concentrations (all tested); as for the emulsification index, the biosurfactant presented thermal, pH and saline stability, in the range of 5°C to 100°C, 4 to 8 and in the concentration of 5%, respectively. The biosurfactant produced showed anionic behavior with a content of 55% of carbohydrates, and
yield of 2.174 g/L. The fungal biomass produced showed a yield of 27.4 g/L and a total lipid concentration of 39.5%. From the selection of the optimized conditions, the co-culture with the microalgae will be investigated, evaluating different parameters such as aeration, luminosity, pH and temperature. Other parameters will be evaluated for the accumulation of lipids in the biomass, during growth and production of metabolites. Stability and toxicity of secondary metabolites produced will also be analyzed. The final stage of the research consists in the extraction and characterization of secondary metabolites produced by co-cultivation with a view to future potential as a supplement for the food industry.
