SUMMARY:

Amazona sp. are the second most trafficked species in Brazil, compromising its conservation
status and enhancing the transmission and mutation of pathogens, posing a risk for zoonotic
spillover. The aim of this study was the phenotypic and genotypic investigation of Extended
Spectrum Beta-Lactamase (ESBL) and Diarrheogenic virulence (DEC) and biofilm production
in Escherichia coli isolated from the intestinal microbiota of birds of the genus Amazona sp.
from the Wildlife Screening and Rehabilitation Center Tangara (CETRAS Tangara), Recife,
Pernambuco. A total of 64 birds of the species Amazona aestiva and A. amazonica were
evaluated and sampled. The clinical sign of diarrhea was found exclusively in animals in
Residence, being present in 43.8% (28/64). A total of 28 E. coli isolates were recovered from
the samples. The isolates were subjected to biofilm production assay, double disk synergism
test (DDST) and molecular diagnosis by PCR of the ESBL genes blaTEM-1, blaSHV-1 and
CTX-M825 and DEC virulence genes eaeA, HlyA, stx1 and stx2. In the phenotypic tests, 100%
(28/28) of the isolates produced biofilm, and 32.1% (9/28) were positive in the DDST for
ESBL production. In the genotypic tests, 13% (4/28) were positive for blaTEM-1 genes. The
blaSHV and CTX-M825 genes were not detected in the analyzed samples. The eaeA gene was
detected in 13% (4/28) of the isolates, without detection of the other genes, confirming the presence of EPEC-type DEC among the investigated animals. Five isolates were positive for two or more phenotypic and genotypic tests and three of these had genotypes for ESBL and EPEC. It is concluded that Amazona aestiva and A. amazonica should be used as environmental bioindicators for Extended Spectrum Beta-lactamase-producing E. coli (ESBL- EC), virulence and biofilm production in Wildlife Screening and Rehabilitation Centers.
