EFFECT OF MELATONIN ON TESTICULAR HISTOPHYSIOLOGICAL PARAMETERS AND METABOLISM IN ADOLESCENT RATS TRAINED AND SUPPLEMENTED WITH NANDROLONE AND/OR CREATINE

SUMMARY:

The indiscriminate use of anabolic steroids (AS) and creatine by young individuals poses a risk to reproductive health. This study aimed to evaluate the effects of the combination of nandrolone decanoate (ND) and creatine on the testes of trained adolescent rats, as well as the protective potential of melatonin. Wistar rats (40 days old) were divided into three trained (swimming) groups: Control (CG); Nandrolone+Creatine (CGNC); and Nandrolone+Creatine+Melatonin (CGNC+Mel). Treatment lasted 6 weeks with DN (5 mg/kg), creatine (0.5 g/kg/day) and/or melatonin (200 µg/100g). Histomorphometric parameters, organosomatic index, oxidative stress (TBARS, GSH) and immunohistochemical markers (VEGF, PCNA, AR) were analyzed. The GTNC group showed severe degeneration of the seminiferous epithelium, testicular atrophy, reduction in tubular diameter, epithelial height and number of sperm, Sertoli and Leydig lineage cells. Increased lipid peroxidation (TBARS), glutathione (GSH) depletion and decreased expression of VEGF, PCNA and AR were also observed. In contrast, melatonin administration (GTNC+Mel group) prevented histopathological changes, normalized oxidative stress markers, and restored morphometric parameters and protein expression to levels similar to those of the control group. Co-administration of nandrolone decanoate and creatine induced acute testicular toxicity in trained adolescent rats, mediated by oxidative stress. Melatonin demonstrated a potent protective effect, mitigating damage and preserving the morphofunctional integrity of the testes
