SUMMARY:

The main objective of this work was to investigate the electrocatalytic oxidation of 7-methylguanine (7-mGua) and 5-methylcytosine (5-mCyt) in different aqueous electrolytes, on the surface of the cathodically pre-treated boron-doped diamond electrode (red-BDDE) and for 3-methyladenine (3-mAde) on the anodically pre-treated glassy carbon electrode (oxi-GCE). Furthermore, we sought to develop a simple, sensitive and selective electrochemical method for the simultaneous quantification of methylated bases, 7-mGua and 5-mCyt, using the BDDE-network and differential pulse voltammetry (DPV), in the presence of possible interferences and for future applications in hydrolyzed DNA samples. An electroanalytical method using DPV and oxi-GCE was also investigated for detection and quantification of 3-mAde. This approach represents a significant innovation in DNA methylation analysis, showing promise for future applications in medical research and diagnosis.
