SUMMARY:

Depression is considered one of the main causes of morbidity and is associated with increased mortality. Its treatment generally follows pharmacotherapy, with the use of antidepressants, which act on the central nervous system (CNS). Among the antidepressants sold, fluoxetine (FLX) has been widely used to treat depression and some disorders. However, despite its effectiveness, many side effects are noted, including suicidal ideation, which has been the focus of substantial concern. Another factor that is considered a serious public health problem is obesity, and therefore the search for alternative treatments, such as the use of dietary supplements, has been recurrent. However, these products are often adulterated, mainly with anorectics, such as amphetamines, which are psychoactive substances with a stimulating action on the CNS and have a high potential to generate dependence. Therefore, the present work aims to investigate through voltammetric techniques, such as cyclic voltammetry (CV), square wave voltammetry (SWV) and differential pulse voltammetry (DPV), using a glassy carbon electrode (GCE) and a boron-doped diamond electrode (BDDE), the electrochemical behavior of the antidepressant FLX in order to establish its redox reactions and stability and a simple, sensitive, selective and rapid electroanalytical method for its detection and quantification in real drug samples. As well as developing a chromatographic method for the simultaneous detection of adulterants, such as amphetamines, in food supplements. Voltammetric results demonstrated that, at GCE, FLX undergoes irreversible electro-oxidation in a single step, which occurs with the transfer of two electrons, with the formation of one electroactive oxidation product. Using BDDE, the presence of a single irreversible oxidation process was verified, associated with the electro-oxidation of FLX. Furthermore, electroanalytical methodologies using BDDE, as well as deoxyribonucleic acid double helices (dsDNA) modified BDDE, for detection and quantification of FLX were investigated and developed, and the limits of detection (LOD) and quantification (LOQ) were calculated and compared. Gas chromatography coupled to mass spectrometry (GC-MS) was used to evaluate the chemical profile of amphetamines, such as methamphetamine, 3,4-methylenedioxymethamphetamine (MDMA), 3,4-methylenedioxyethylamphetamine (MDEA) and 3,4-methylenedioxyamphetamine (MDA), with the aim of detecting and quantifying these species, in food supplements, in order to solve problems in the field of forensic chemistry. Different conditions were also evaluated in order to optimize the proposed method, such as the effect of varying concentration, pH of the medium, solvent and the use of derivatizers.
